 We continue our lesson on Microscopic from where we stop previous class. We learn about how to prepare a specimens for microscopy. Each different microscope require the specimen to be prepare by different way. For light microscope the preparation steps include wet mount and smears. Smears step involve preparing smears, fixation and staining. There are many type of staining. Every microorganism use different stain and each of strain has its function.

    I personally was shock to know that there are many types of stain. I before this only see slide prepared using microscope. I do not expect to make this slide it is very complex. The microorganism details will be lost if we use wrong stain and wrong technique. That means we have to appreciate the slide being prepared for us during our school day because of hardship to make the slide and I also have to learn correct way on preparing slide which being use by me during studying microbiology.
     A bit more information about staining we learn during class. 
  • Gram positive staining  is only for bacteria
  • Malachite green is an active reagent used for staining protozoa which always be said in class
This video was showed during class really help me understand better about grand stain. It help me gets the idea of Grain Stain.

 Mycobacterium is a fussy bacteria which cause Tuberculosis. The process of preparing slide for mycobacterium must be do it with correct stain and correct technique.



At the end of the class we have to do an online mind map about External prokaryote structure which the topic for next class.

What have I learn:
-Gram positive staining is only for bacteria
-In staining  Protoza malachite green is used
-Some microorganism very fussy.
